Agaricus blazei Polypeptide: Sourcing the beta-Glucan Fraction

A mushroom-derived polypeptide fraction is bought on two claims that pull in opposite directions. The buyer wants a protein-rich hydrolysate, but the mushroom’s commercial identity rests on its cell-wall polysaccharides and its sterols. A supplier who over-purifies the protein delivers a material that assays well and no longer tests like a mushroom. Sourcing Agaricus blazei polypeptide therefore means specifying what must be retained, not only what must be high.

The cell-wall polysaccharide: the 162.14 g/mol rule

The beta-glucan of fungal cell walls is catalogued under CAS 9041-22-9. PubChem (CID 439262) lists the glucan segment as C18H32O16 at 504.4 g/mol, which is a three-residue piece of chain: 3 × 180.16 − 2 × 18.02 = 504.44 g/mol. Each further glucose residue in the chain adds 180.16 − 18.02 = 162.14 g/mol. That constant is the whole basis of a beta-glucan assay sanity check: a “beta-glucan content” figure expressed as a percentage of dry mass is only meaningful when the method states which linkage range it recognises, because a branched beta-1,3/1,6 chain and a linear beta-1,3 chain differ in residue arrangement while sharing the same 162.14 g/mol increment.

The sterol marker: ergosterol and a mass-neutral photochemistry

Ergosterol, CAS 57-87-4, C28H44O, 396.6 g/mol (CID 444679) is the fungal sterol, and it is the single most useful identity marker in a mushroom material because plants do not make it at this level. Under ultraviolet exposure it undergoes ring opening to previtamin D2 and then to ergocalciferol. The point worth noting for a spec sheet: that photochemical opening is mass-neutral. Ergosterol, previtamin D2 and vitamin D2 all sit at C28H44O and 396.6 g/mol, so a mass-based method cannot distinguish them and an ultraviolet-treated lot will pass a sterol assay while having a completely different biological profile.

The soluble carbohydrate pair

Trehalose, CAS 99-20-7, C12H22O11, 342.30 g/mol (CID 7427) is the fungal disaccharide: two glucoses joined with both anomeric carbons locked, so 2 × 180.16 − 18.02 = 342.30 g/mol and the molecule is non-reducing. Mannitol, CAS 69-65-8, C6H14O6, 182.17 g/mol (CID 6251) is the polyol. Both are small and water-soluble, and both will inflate a non-specific carbohydrate result unless the method precipitates the polymer first. A supplier quoting a polysaccharide percentage without excluding them is reporting something closer to total soluble solids.

The peptide side of the material

On the protein side the same residue arithmetic applies as to any hydrolysate: a free amino acid loses 18.02 g/mol on incorporation. Glycine at 75.07 g/mol becomes 57.05 g/mol as a residue, glutamic acid at 147.13 g/mol becomes 129.11 g/mol, aspartic acid at 133.10 g/mol becomes 115.08 g/mol, and lysine at 146.19 g/mol becomes 128.17 g/mol. A certificate that gives an amino acid profile should be read in residue terms; comparing it against a free-amino-acid calibrated standard will not close to 100%.

What to put in the purchase specification

RequirementWhy it belongs in the contract
Name the beta-glucan method and linkage rangeBranched and linear chains share the 162.14 g/mol increment
Set a minimum ergosterol, not just a sterol totalErgosterol at 396.6 g/mol is the fungal marker; phytosterols are not
Exclude trehalose and mannitol from the polymer figure342.30 and 182.17 g/mol species inflate total carbohydrate
State whether UV treatment was appliedRing opening is mass-neutral and invisible to a sterol assay
Report the amino acid profile in residue termsEach residue is 18.02 g/mol lighter than the free acid

Frequently asked questions

Is a higher beta-glucan number always better?

Not if it was achieved by stripping the protein. The polypeptide claim and the polysaccharide claim come from different fractions of the same fruiting body and a spec should floor both.

Why does the ergosterol result look unchanged after UV treatment?

Because ergosterol, previtamin D2 and vitamin D2 all carry C28H44O at 396.6 g/mol; the conversion changes shape, not mass.

Can trehalose be used as an identity marker?

It supports one. At 342.30 g/mol and non-reducing, it is characteristic of fungi, but it is also widely used as a food ingredient, so it must be read together with ergosterol.

Identity data in this article is taken from PubChem (US National Library of Medicine) records for CIDs 439262, 444679, 7427, 6251, 750, 33032, 5960 and 5962. Batch-specific values – assay, moisture, particle size, packaging, shelf life and minimum order quantity – are issued per lot on the certificate of analysis and are not published here.
